Mutation resulted in the ablation of specific binding. Labeling was observed in permeabilized preparations using an antibody specific for the N terminus of the 5-HT3A subunit.
Because of the cis nature of this proline, its adjacent residues were also mutated: mPhe169 and mPhe171 to alternative "favorable" and "non-favorable residues."
